YILIMART - Pan DC Isolation Kit, mouse

The Pan Dendritic Cell Isolation Kit, mouse is a fast enrichment kit for untouched isolation of all dendritic cell subpopulations, both classical dendritic cells and plasmacytoid dendritic cells, from murine spleen.

Detailed separation procedure

Using the Pan Dendritic Cell Isolation Kit, mouse dendritic cells are isolated by depletion of non-target cells (negative selection). Non-target cells are labeled with a cocktail of biotin-conjugated monoclonal antibodies against antigens that are not expressed by dendritic cells. In a second step, cells are labeled with Anti-Biotin MicroBeads. The magnetically labeled non-target cells are depleted by retaining them within a MACS Column in the magnetic field of a MACS Separator while the unlabeled DCs run through.

Downstream applications

Dendritic cells isolated with the Pan Dendritic Cell Isolation Kit can be used for:

Analysis of antigen uptake, processing, and presentation
Stimulation of primary T cell responses by antigen-pulsed dendritic cells
Elucidation of specific signal transduction pathways

Pan DCs were isolated from mouse spleen cell suspension using the Pan Dendritic Cell Isolation Kit, mouse, an LS Column, and a MidiMACS™ Separator. The cells were fluorescently stained with CD11c-APC and mPDCA-1-FITC and analyzed by flow cytometry using the MACSQuant

Analyzer. Cell debris, dead cells and lineage positive cells were excluded from the analysis based on scatter signals, propidium iodide fluorescence and lineage antigen expression.
