The CD115 MicroBead Kit has been developed for positive selection of CD115

mouse cells from lymphoid tissues.

Background information

CD115 is expressed on monocytes, macrophages, osteoclasts, as well as on common dendritic cell precursors and macrophage/dendritic cell precursors. CD115 has been proposed as a marker for the standardized identification and analysis of mouse monocytes.

Detailed separation procedure

In order to avoid non-specific binding of the antibody, the single-cell suspension is pre-incubated with FcR Blocking Reagent. Target cells are then labeled with a biotinylated CD115 antibody. Subsequently, CD115

⁺

cells are magnetically labeled with Anti-Biotin MicroBeads. After the separation over a MACS Column, the CD115

⁺

cells are eluted as the positive fraction.

Downstream applications

Isolated mouse CD115

⁺

cells , e.g. from bone marrow, can be used for

in vitro

analysis, differentiation studies, co-culture, or adoptive transfer experiments.

CD115

⁺

cells were isolated from C57BL/6 bone marrow cells using the CD115 MicroBead Kit, two MS Columns and an OctoMACS Separator. Cells were fluorescently stained with CD115-PE and Anti-Ly-6C-APC and analyzed using the MACSQuant® Analyzer. Cell debris and dead cells were excluded from the analysis based on scatter signals and propidium iodide fluorescence.
