YILIMART - Anti-Ly49A/D-FITC, mouse, REAL400, 100 t

Clone REAL400 is an antibody fragment derived from the full Anti-Ly-49A/D antibody molecule. It displays no binding to Fc receptors. The recombinantly engineered antibody fragments are multimerized to form the REAlease Complex to bind markers with high avidity.

Clone REAL400 recognizes the mouse Ly-49A and Ly-49D antigens belonging to the Ly-49 family. Members of Ly-49 family are homodimeric C-type lectin receptors, recognizing MHC class I molecules as ligands, and include activating and inhibitory receptor molecules. Ly-49A is a 44 kDa type II transmembrane, disulfide-linked homodimer with C-type lectin extracellular domains. Expression of Ly-49A is found on subset of natural killer (NK) and NK-T cells. Ly-49D is a prototypic activating member of Ly-49 family and in contrast to inhibitory members, lacks a cytoplasmic immunoreceptor tyrosine-based inhibitory motif (ITIM). A positive signal is initiated by interaction of Ly-49D with immunoreceptor tyrosine-based activating motif (ITAM) containing transmembrane adapter DAP12. Clone REAL400 does not react with BALB/c, DBA/2, AKR, or CBA/J Ly-D49.

The REAlease Kits consist of the respective fluorochrome-conjugated REAlease Complexes and the REAlease Support Kit for removal of the REAlease Complexes and optional relabeling with different fluorochrome-conjugated REAlease Complexes.

Splenocytes from C57BL6 mice were stained with REAlease Anti-Ly-49A/D-FITC (B) or with the corresponding REAL Control (A). REAlease Anti-Ly-49A/D-FITC was released using the REAlease Release Reagent (C). To show epitope accessibility after REAlease Complex release, the cells were washed with the REAlease Stop Reagent and restained with REAlease Anti-Ly-49A/D-FITC (D). Cells were labeled with Anti-NK1.1 antibodies in addition and analyzed by flow cytometry using the MACSQuant

Analyzer. Cell debris and dead cells were excluded from the analysis based on scatter signals and propidium iodide (PI) fluorescence.
