GMP CD8-APC consists of a mouse monoclonal antibody against the CD8 antigen conjugated to a fluorochrome. The CD8 antibody (clone BW135/80) recognizes the CD8 antigen, which is a disulfide-linked dimer, existing either as a CD8 α homodimer or as a CD8 α/β heterodimer.

Background information

Clone BW135/80 recognizes the human CD8 antigen. This cell surface glycoprotein is present on 20-30% of human peripheral blood lymphocytes. CD8 is strongly expressed on CD8-positive T cells and thymocytes and dimly on a subset of NK cells and CD8-positive γ/δ Τ cells. CD8 acts as a co-receptor with MHC class I-restricted T cell receptors in antigen recognition.

GMP CD8 Fluorescent Antibodies are for identification of CD4-positive cells by flow cytometry as well as cell enrichment by flow cytometric sorting.

A leukapheresis sample was labeled with MACS

GMP CD8-APC and with CD3-VioGreen for gating purposes. Cells were analyzed by flow cytometry using the MACSQuant Analyzer 10. Cell debris and dead cells were excluded from the analysis based on scatter signals and propidium iodide fluorescence. Viable lymphocytes were further gated on CD3

cells.
