Whole Blood Eosinophil Isolation Kit has been developed for the fast isolation of untouched eosinophils from freshly drawn anticoagulated whole blood without density gradient centrifugation and red blood cell lysis. Sample volumes of up to 30 mL can be processed.

Detailed separation procedure

With the new MACSxpress

^®

Technology, untouched human leukocyte subsets can be isolated from up to 30 mL of anticoagulated whole blood. Non-target cells are removed by immunomagnetic depletion using MACSxpress Beads. Simultaneously, erythrocytes are sedimented, yielding untouched target cells of high purity.

Whole Blood Eosinophil Isolation Kit, a MACSmix™ Tube Rotator, and a MACSxpress Separator. The isolated cells were fluorescently stained with Anti-Siglec-8-APC, CD45-VioBlue

and CD15-PE and analyzed by flow cytometry using the MACSQuant

Analyzer 10. Cell debris, non-leukocytes, and dead cells were excluded from the analysis based on CD45 expression, scatter signals, and propidium iodide fluorescence.
