Whole Blood NK Cell Isolation Kit was developed for the isolation of untouched natural killer (NK) cells from anticoagulated whole blood without density gradient centrifugation.

Detailed separation procedure

With the new MACSxpress

Technology, untouched human leukocyte subsets can be isolated from up to 30 mL of anticoagulated whole blood. Non-target cells are removed by immunomagnetic depletion using MACSxpress Beads. Simultaneously, erythrocytes are sedimented, yielding untouched target cells of high purity.

Whole Blood NK Cell Isolation Kit can be used, e.g., for phenotyping, NK cell expansion or cytotoxicity assays.

Untouched NK cells were isolated from 30 mL of human EDTA anticoagulated whole blood using the MACSxpress

Whole Blood NK Cell Isolation Kit, a MACSmix Tube Rotator, and a MACSxpress Separator. The isolated cells were fluorescently stained with CD45-VioBlue

, CD3-FITC, and CD56-PE and analyzed by flow cytometry using the MACSQuant

Analyzer 10. Cell debris, non-leukocytes, and dead cells were excluded from the analysis based on CD45 expression, scatter signals, and propidium iodide fluorescence.
